Acetylchlorid wird durch die Umsetzung von Calciumchlorid mit Essigsäureanhydrid hergestellt. Alternativ lässt es sich durch Umsetzung von Essigsäure mit Phosphor(III)-chlorid erhalten. Die Synthese aus Essigsäure und Thionylchlorid bzw. Phosphor(V)-chlorid ist ebenfalls möglich, liefert aber schlechtere Ausbeuten.
