Study of the activity of the VIN 11 extract

Rakotoarimalala Nomembalisoa Tahiry
The objective of this work was to study the effect of VIN 11 extract on diarrhea in guinea pigs. Its anti-secretory effect was studied in vivo on intestinal hypersecretion induced by 2 g/kg of magnesium sulfate, administered orally. While its effect on intestinal hypermotility was studied in vitro on isolated duodenum contracted with acetylcholine. Our results show that VIN 11 extract decreases intestinal hypersecretion induced by magnesium sulfate. The volume of intestinal fluid in controls was equal to 5.82 ± 0.68 ml, compared to 3.6 ± 0.28, 1.87 ± 0.36 ml and 0.85 ± 0.14 ml for animals treated with the extract at doses of 100, 200 and 400 mg/kg (P<0.05). In addition, VIN 11 extract inhibits acetylcholine-induced intestinal motility, with an EC50 equal to 0.7 mg/ml, and inhibits the action of acetylcholine in a non-competitive manner. In the absence of the extract, the maximum contraction of the duodenum caused by acetylcholine is equal to 50% with an EC50 of 9.10-9 M.
